The phrase "player your club can least afford to lose" can be interpreted in many different ways. It doesn't mean best player or most influential all the time.
There are multiple factors why a team couldn't afford to lose a certain player, such as depth in their position, how much they influence the game plan or whether they could fill the void more easily than others.

Brisbane
The back-to-back premiers have a big list of stars, but co-captain Harris Andrews would be the hardest to replace if he were to go down, rather than some of their midfield stars.
Andrews is the leader of their backline and has consistently held up the Lions' defence over the years. With the struggles the Lions are currently having in defence, giving up 18 more points this year than last. Missing Andrews would make this look even worse than what it already is.
