Melbourne lost recruit and key forward Brody Mihocek to concussion in Friday's match simulation, which could put him in doubt for Round 1.
Mihocek won a dangerous tackle free kick against North Melbourne's Charlie Comben, although it hasn't been confirmed that the incident was the cause of the injury.
The 33-year-old had been working back from an interrupted pre-season, but this latest setback rules him out of next week's practice match against Richmond.
Mihocek is one of multiple players entering concussion protocols in the past week, with Liam Duggan, Jack Silvagni, Ryan Lester, and Oscar Allen set to miss their respective club practice matches.
The key forward crossed from Collingwood in the pre-season, and had impressed in his new colours in the first quarter, slotting two goals.

North Melbourne defender Riley Hardeman has sent fears into Arden Street after he fell and clutched at his left ankle in Friday's match simulation.
During the second term, Hardeman contested a mark against Melbourne's Tom Sparrow, but immediately pulled up lame.
The 20-year-old was assisted from the field by Kangaroos trainers, but was showing signs that he couldn't put weight on his left leg.

Hardeman has been ruled out of the remainder of the clash with an ankle injury.
